Basically both operate by bouncing tiny particles or waves off of objects in order to make them visible. Electrons are smaller than light photons so we can see smaller objects clearly with electron microscopes. There are disadvantages though. First, color is a function of light wave frequency so we can't see electron micrographs in color. Color can be added artificially though by computer to differentiate structures, (think photoshop). Also, we cannot see electrons so we make them visible by bouncing them off a cathode ray screen similar to a TV set or computer monitor :D

The absolute basic thing you have to know though is that with a light microscope, you're only limited to being able to see a certain size due to the fact that you're using light to illuminate your objects, and visible light has wavelengths between ~400 and ~700 nanometers. WIth electron microscopes, you're not limited to this so you can see much smaller things.
A third (practical) difference is that preparing a sample for an electron microscope kills it, so it is impossible to view live things under it. For observing actual living processes, you need a light microscope. Not as theoretically elegant as spatcher's answer but, depending on what you are trying to look at, also important.
